Synthesis and evaluation of fosfomycin group end-capped packing materials for hydrophilic interaction liquid chromatography

Hydrophilic interaction liquid chromatography (HILIC) plays an important role in the analysis of compounds having high polarity. In this study, fosfomycin (F) was chosen as a new end-capping reagent, owing to facile hydrolysis reaction its epoxy group. Zirconia coated silica (ZrO2/SiO2) materials good chemical and physical stability were prepared. D-glucose-6-phosphate (G) D-fructose1,6-bisphosphate (FDP) modified onto inner outer surfaces ZrO2/SiO2 microbeads. The reagent then bonded surface material through Lewis acid-base interactions. properties (morphology, Zr content, pore size, volume, carbon content) stationary phases (SPs) characterized. Finally, resulting end-capped SPs employed separate alkaloids benzoic acids. Multiple interactions, including HILIC, electrostatic repulsion, ion exchange hydrogen bonding, contributed retention analytes on SPs. On F-FDP-ZrO2/SiO2 column, theoretical plate number 31,700 plates/m asymmetry factor 1.63 achieved for berberine, exhibiting chromatographic performance. Furthermore, FDP-ZrO2/SiO2 column showed potential acid Sprite ginsenoside separations. Thus, results indicated significant using F reagent.
